elisa试剂盒总需要时间来保温，是因为ELISA属固相免疫测定，抗原、抗体的结合只在固相表面上发生。以抗体包被的夹心法为例，加入板孔中的标本，其中的抗原并不是都有均等的和固相抗结合的机会，只有zui贴近孔壁的一层溶液中的抗原直接与抗体接触。这是一个逐步平衡的过程，因此需经扩散才能达到反应的终点。

保温的方式除有的ELISA仪器附有特制的电热块外，一般均采用水浴，可将ELISA试剂盒板置于水浴箱中，ELISA板底应贴着水面，使温度迅速平衡。为避免蒸发，板上应加盖，也可用塑料贴封纸或保鲜膜覆盖板孔，此时可让反应板漂浮在水面上。若用保温箱，ELISA试剂盒应放在湿盒内，湿盒要选用传热性良好的材料如金属等，在盒底垫湿的纱布，zui后将ELISA板放在湿纱布上。湿盒应先放在保温箱中预温至规定的温度，特别是在气温较低的时候更应如此。无论是水浴还是湿盒温育，反应板均不宜叠放，以保证各板的温度都能迅速平衡。室温温育的反应，操作时的室温应严格限制在规定的范围内，标准室温温度是指20～25℃，但具体操作时可根据说明书的要求控制温育。室温温育时，人高尔基体抗体(AGAA)ELISA试剂盒只要平置于操作台上即可。应注意温育的温度和时间应按规定力求准确。为保证这一点，一个人操作时，一次不宜多于两块板同时测定。
