军团菌的认识
1976年国费城退伍军人协会会员中曾爆发急性发热性呼吸道疾病,经研究,发现一种细菌命名为嗜肺军团菌。随后,许多有关细菌暂被列入这一属,且追溯研究发现早在1943年即有军团人员病的病例。现已提出了超过30种军团杆菌,至少19种是人类肺炎的病原。其中zui常见病原体为嗜肺军团菌(占病例的85%~90%),其次是L。micdadei(占5%~10%),再次是L。bozemanii和L。dumoffii。此类细菌形态相似,具有共同的生化特征,引起类似疾病。
嗜肺军团菌则暴发流行,军团病可发生于任何年龄,但大多数人都是中年男性。已确定的危险因素包括吸烟,滥用酒精和免疫抑制,特别是由皮质类固醇引起的免疫抑制。
2010年7月12日,英国健康防护局的研究发现,司机罹患退伍军人症的几率是一般人的5倍,主因可能是,车辆的雨刷水箱加一次水,往往要很多天才会用完,箱底常留有残水,因此易变成孳生退伍军人杆菌的温床。而据英国的统计,约有超过20%的退伍军人症病例,是通过汽车雨刷水箱感染的。

以急性发热性呼吸道症状为特征的退伍军人症是由吸入含有嗜肺军团菌的雾化水引起的。嗜肺军团菌血清组1zui常从患者中分离(1,2)。
IMMUNOCATCHTM军团菌(Legionella)是一种侧流免疫层析法,可通过快速可靠地检测尿样中的抗原,对嗜肺军团菌血清型1感染进行早期诊断。
原理
本产品是一种试剂盒,设计用于免疫层析法检测尿样中嗜肺军团菌血清型1 LPS抗原。
当样品通过毛细作用迁移通过结合垫时,样品中的军团菌抗原与缀合至胶体颗粒的抗军团菌抗体结合形成抗原 - 抗体复合物。抗原 - 抗体复合物在试纸条上迁移,与固定在膜上的抗军团菌抗体特异性结合而产生红线。然后通过肉眼观察红线的存在来确定军团菌抗原的存在。
此外,抗-Legionella抗体偶联的胶体金在反应中不被消耗,与固定在对照线上的抗免疫球蛋白抗体结合产生红线,表明在测试条上的反应是成功的。

【研发原理】
军团菌是近年来发现的新型病原菌,它侵入人体后主要引起军团菌肺炎。据发达国家报道,军团菌肺炎占肺部感染总数的7%~10%,老年人中会更高。该病发病急、死亡率高,因此临床上迫切需要能够快速准确检测军团菌的方法。根据市场需要,我公司攻克多项技术难关,成功研制出军团菌抗体快速金标记检定卡。

【检测方法】
1)从铝袋中取出所需数量的磁带,并将它们放在水平面上。
2)使用定量移液器慢慢地吸取90μL或更多的样品。 通过一次移液操作,将全部样品滴入盒子的样品位置。 滴下后,剩余的标本留在吸管内。
3)在室温(15-30°C)下将盒子放置15分钟,
4)检查盒子评估地点是否有线(标有“C”和“T”)。
结果
应该在反应开始15分钟后及时进行评估。 检查评估地点,并通过下面描述的红线来评估结果。
1)正面:红色控制线和测试线均在评估现场。 当两条线在反应时间结束之前出现时结果是肯定的。
2)否定的:评估地点只有一条红色的控制线。
3)无效:无论控制线是否存在,无论是否存在测试线,测试均无效。

The pathogenesis of dengue haemorrhagic fever has three hypotheses: First, the virulence of the virus strains are different. Type II viruses cause dengue hemorrhagic fever, other types of viruses cause dengue fever. Second, the virus mutation, that the virus gene mutation led to increased virulence, but the evidence of the current virus mutation is not sufficient. Third, the theory of secondary infection, that the first infection of any type of dengue virus, only mild or typical dengue occurred, and when the second infection, no matter what type of virus, which is manifested as dengue haemorrhagic fever. Some 55 cases of dengue haemorrhagic fever were reported, 51 of which were secondary infections. The mechanism is that when the second infection, the body memory reaction occurs, resulting in high titers of IgG, with the antigen to form immune complexes. Binds to large monocytes or macrophage surface Fc receptors, and activates these cells to release proteases, thromboplastins and vascular permeability factors that cleave complement C3. These enzymes and factors activate the complement system and the coagulation system, resulting in increased vascular permeability and exudation of plasma proteins and blood components, causing pathological changes such as blood concentration, bleeding and shock.
Pathological changes of the liver, kidney, heart and brain degeneration; endocardial, pericardial, pleural, gastrointestinal mucosa, muscle, skin and central nervous system of varying degrees of bleeding; rash within the small endothelium swelling, perivascular edema and Monocyte infiltration. Severe patients may have central hepatic lobule necrosis and cholestasis, lobular pneumonia, pulmonary abscess formation. Pathological changes of dengue haemorrhagic fever as systemic microvascular damage, leading to plasma protein oozing and bleeding. Digestive tract, subendocardial, subcutaneous, hepatic subcapsular, pulmonary and soft tissue exudate and bleeding, visceral small blood vessels and microvascular edema, bleeding and lymphocyte infiltration. Brain autopsy showed subarachnoid and intraparenchymal hemorrhage, brain edema and brain softening.
Latency 5 ~ 8d. According to the WHO standards are divided into typical dengue fever, dengue haemorrhagic fever and dengue shock syndrome type 3. Dengue fever seen in our country in recent years can be divided into typical dengue fever, light dengue fever and heavy dengue fever.
