裸鼠环加氧酶2<COX-2>ELISA试剂盒敏感性高、操作简便，配套仪器设备的发展使操作程序规范化和自动化，并进一步提高了稳定性，被广泛用于检测多种病原体抗原或抗体、血液及其他体液中的微量蛋白成分、细胞因子等。ELISA应用的范围很广，而且正在不断地扩大，原则上ELISA可用于检测一切抗原、抗体及半抗原，可以直接定量测定体液中的可溶性抗原。
(1)检查抗原和半抗原方面。尚有用于检查大肠杆菌、铜绿假单胞菌、破伤风梭菌毒素、轮状病毒、呼吸道融合及巨细胞病毒等。在免疫化学方面可用于检测疯牛病、痒病等病原学检测。
(2)检查抗体方面。用ELISA间接法检查抗体，已获得对多种传染病和寄生虫病的血清学诊断，亦开始广泛用于现场流行病学调查。在寄生虫病方面，它用于对阿米巴、血吸虫、肺吸虫、肝吸虫、旋毛虫病等血清学诊断。在病原微生物方面已用于检查链球菌、沙门菌、布氏杆菌、结核杆菌、流感病毒、轮状病毒、疱疹病毒、狂犬病毒等，其敏感性都超过目前常用的检查方法。
分类：ELISA常用的方法主要包括双抗体夹心法、间接法、竞争法以及BAS-ELISA。
(1)双抗体夹心法：双抗体夹心法是检测抗原zui常用的方法。夹心法利用两种一抗对目标抗原进行捕获和固定，在确保灵敏度的同时大大提高了反应的特异性，该方法的检测灵敏度可提高到pg级的水平。将已知抗体包被到固相表面，加入待检标本，标本中若含有相应抗原即与固相表面的抗体结合，洗涤去除未结合成分，加入该抗原特异的酶标记抗体，洗去未结合的酶标记抗体，加底物后显色。若标本中无相应抗原，固相表面即无抗原结合，加入的酶标记抗体则不能结合于固相并被洗涤去除，当加入无色底物后，因无酶催化故不显色。双抗体夹心法适用于测定2价或2价以上的大分子抗原，但不适用于测定半抗原及小分子单价抗原，因其不能形成两位点夹心。
(2)间接法：间接法是检测抗体zui常用的方法，其原理为利用酶标记的抗抗体以检测已与固相结合的受检抗体，故称为间接法。先将待测的蛋白包被在孔板内，然后依次加入一抗、酶标记的二抗和底物显色，通过仪器(例如酶标仪)定量检测抗原。这种方法操作简单但由于高背景而特异性较差，目前已逐渐被夹心法取代。间接法的优点是只要变换包被抗原就可利用同一酶标二抗建立检测相应抗体的方法。间接法成功的关键在于抗原的纯度。间接法中另一种干扰因素为正常血清中所含的高浓度的非特异性抗体。
(3)竞争法：竞争法可用于测定抗原，也可用于测定抗体。当抗原材料中的干扰物质不易除去，或不易得到足够的纯化抗原时，可用此法检测特异性抗体。其原理为标本中的抗体一定量的酶标抗体竞争与固相抗原结合。标本中抗体量越多，结合在固相上的酶标抗体愈少，因此阳性反应呈色浅于阴性反应。如抗原为高纯度的，可直接包被于固相。如抗原中有干扰物质，直接包被不易成功，可采用捕获包被法，即先包被与固相抗原相应的抗体，然后加入抗原，形成固相抗原。洗涤除去抗原中的杂质，然后再加入标本和酶标抗体进行竞争结合反应。小分子抗原或半抗原因缺乏可用于夹心法的两个以上的位点，因此不能用双抗体夹心法进行测定，可以采用竞争法模式。其原理是标本中的抗原和一定量的酶标抗原竞争与固相抗体结合。标本中抗原量含量愈多，结合在固相上的酶标抗原愈少，zui后的显色也愈浅。
