【预期用途】

尼古丁(Nicotine)是烟草中的主要生物碱，是导致吸烟成瘾的物质动因，也是评价人体摄入烟草烟雾的常用指标。但因为尼古丁半衰期短，无法作为标志物检测，其代谢物可替宁因为半衰期长作为吸烟和戒烟的标志物。

本品采用竞争抑制法和胶体金免疫层析技术，用于快速定性检测人体唾液中的可替宁，适用于评价烟草烟雾摄入的初步筛查。

【主要组成成份】

• 可替宁检测试剂盒（40人份）：每人份铝箔袋单独包装。其中试剂盒由金标可替宁单克隆抗体、可替宁-BSA结合物、羊抗兔IgG多克隆抗体、硝酸纤维素膜、聚酯纤维素膜、塑料背衬、塑料模板组成。
• 一次性灭菌唾液取样棒（40人份）
• 唾液收集器（40人份）
• 使用说明书（1份）

• 【检验方法】

• 撕开铝箔袋，取出试剂，应在1小时内尽快使用。
• 拿出一次性唾液收集器。（步骤1）
• 手持收集器盖帽使海绵头含入口中，直至海绵头吸满液体（膨胀变软）。（步骤2）
• 将海绵头放回收集器并拧紧盖帽，使所含样本流入收集器下端。（步骤3）
• 将试剂盒置于干净平坦的台面上，同时打开滴样孔盖帽。（步骤4）
• 挤压收集器上端管壁，垂直滴加2滴无空气泡的唾液（约100ul）于加样孔（S）中。(步骤5)
• 等待紫红色条带的出现，3-5分钟时直接观察结果，10分钟后判定无效。

c)注射方法：先以75％酒精消毒术部，然后皮内注射定量的牛型结核分枝杆菌PPD，注射后局部应出现小疱，如对注射有疑问时，应另选15cm以外的部位或对侧重作。
d)注射次数和观察反应：皮内注射后经72h判定，仔细观察局部有无热痛、肿胀等炎性反应，并以卡尺测量皮皱厚度，作好详细记录。对疑似反应牛应立即在另一侧以同一批PPD同一剂量进行第二回皮内注射，再经72h观察反应结果。对阴性牛和疑似反应牛，于注射后96h和120h再分别观察一次，以防个别牛出现较晚的迟发型细菌反应。4.1.1.2 结果判定
4.    a) 阳性反应：局部有明显的炎性反应，皮厚差大于或等于0mm。
5.    b) 疑似反应：局部炎性反应不明显，皮厚差大于或等于0mm、小于4.0mm。
6.    c) 阴性反应：无炎性反应。皮厚差在0mm以下。
凡判定为疑似反应的牛只，于*次检疫60d后进行复检，其结果仍为疑似反应时，经60d再复检。如仍为疑似反应，应判为阳性。4.1.2 其他动物牛型结核分枝杆菌PPD皮内细菌反应试验
参照牛的牛型结核分枝杆菌PPD皮内细菌反应试验进行。
4.2 禽型结核分枝杆菌PPD皮内细菌反应试验
禽型结核分枝杆菌PPD皮内细菌反应试验是zui广泛使用于家禽的试验。牛及其他哺乳动物也可用牛型结核分枝杆菌PPD和禽型结核分枝杆菌PPD同时在不同的部位进行，以区分特异性和非特异性细菌反应。
4.2.1 禽的禽型结核分枝杆菌PPD皮内细菌反应试验
4.2.1.1 操作方法用10mm×0.5mm针头肉垂皮内注射0.1mL(0.25万IU)禽型结核分枝杆菌PPD，48h后观察结果。4.2.1.2 结果判定
阳性反应为接种部位肿胀，从5．0mm直径的小硬结到扩展至其他肉垂与颈部的广泛性水肿。火鸡肉垂的结核分枝杆菌PPD皮内细菌反应试验，没有家禽的试验那样可靠。其他禽类也可用翼蹼作试验，但结果一般不理想。水禽可接种脚蹼，但试验不敏感并经常与接种部位的感染相混淆。

Where the determination of the suspected reaction of cattle, 60d after the first quarantine for re-examination, the result is still suspected reaction, re-examination after 60d. 4.2 avian Mycobacterium tuberculosis PPD intradermal bacterial response test
Avian Mycobacterium tuberculosis PPD The intradermal bacterial response test is the most widely used test for poultry. Cattle and other mammals may also be used with M. bovis PPD and M. avium PPD simultaneously in different locations to distinguish between specific and nonspecific bacterial reactions.
